The cover is of a rainbow pattern, with the title logo colored in a rainbow pattern continuing from the previous volume's pattern. The author's name is yellow. The cover features Monkey D. Luffy in the middle, Portgas D. Ace and Yamato in the background, and Sanji and Roronoa Zoro in the foreground.

Author's Notes "It sounds like there are some upset people online. I'm sorry if anyone says anything to you at school." "I'm fine!!" says the little guardian. Big guardian: "Cheer up!!" A bunch of delicious food appears!! "I'm so exhausted..." "Get in!!" Car drives off!! Arrive at the hot spring!! "I caught a cold..." "Hey, that's not a cold!!" Off to check in at the hospital!! I'm the one who gets all the glory, but when you're in the vicinity of a famous person, there are lots of bad things that happen too. If I didn't have these bright guardian spirits around, I can tell you that I'd have collapsed long ago!! May I use the milestone of the 100th volume to express my appreciation to them? Thank you for everything, now and always!! It's volume 100!! Hope you enjoy it!!!

• 1005. Devil Child (悪魔の子, Akuma no Ko^?): Robin and Brook save Sanji from Black Maria and confront the Tobiroppo.
• 1006. The Honorable Hyogoro of the Flower (侠客“花のヒョウ五郎”, Kyōkaku "Hana no Hyōgorō"^?): Having regained his youthful strength while on the verge of dying from the Ice Oni, Hyogoro overwhelms the Beasts Pirates.

• 1010. Color of the Supreme King (覇王色, Haō-shoku^?): Luffy realizes he can imbue his body with Supreme King Haki and uses it to clash with Kaidou alone as the other Supernovas retreat.
• 1011. The Code of Anko (あんこの仁義, Anko no Jingi^?): Big Mom enters the Skull Dome, where she reunites with Tama and knocks out Page One after learning what the Beasts Pirates did to Okobore Town.
• 1012. Itch (うず, Uzu^?): A three-way battle ensues between Tama's group, Ulti, and Big Mom.
• 1013. Anarchy In The B.M. (Anarchy（アナーキー） In（イン） The（ザ） BM（ビッグ・マム）, Anākī In Za Biggu Mamu^?): Big Mom takes down Ulti, but is confronted by Kid before she can attack Tama's group.
• 1014. Life's Lousy Actor (人生の大根役者, Jinsei no Daikon Yakusha^?): Kaidou defeats Luffy and heads into the Skull Dome to find Momonosuke. Kanjuro finds the child first, but is struck down by Kin'emon.
• 1015. Chains (縁（くさり）, Kusari^?): Kaidou finds Momonosuke's group and takes down Kin'emon, but is then confronted by Yamato.

• It is revealed that Black Maria has tampered with her Human-Beast Form via the use of drugs. Oda also provided a rough illustration of how she originally looked.

• As a special event for the 100th volume, Eiichiro Oda produced a three volume cover spread encompassing volumes 99, 100 and 101.
  • This was the second time that Oda created a volume cover spread. The first one was with volumes 21 and 22.
  • Volume 13 of One Piece Magazine included a full spread illustration of the three volume covers.
• The release of this volume was commemorated with a series of events and publications under the tagline One Piece 100 We Are ONE.
