The DEA was first mentioned in Grand Theft Auto: Vice City in a recorded audio of Ricardo Diaz by the Vice City Bureau of Investigation in the official GTA Vice City website.
Grand Theft Auto: San Andreas[]
The dossier carrier in the mission Misappropriation is mentioned by Frank Tenpenny to be a DEA agent in the opening cutscene. Carl Johnson kills this agent during the mission.
Grand Theft Auto: Vice City Stories[]
They appear in the mission The Exchange for the deal that was made by Ricardo Diaz. One of the agents appears in the mission The Mugshot Longshot to get Jerry Martinez into the witness protection program. And they were mentioned in the mission Turn on, Tune in, Bug out when they bugged Lance's apartment in order to collect some evidence against the Vance brothers to have them arrested. They also sometimes monitor the handover of drugs during Drug Runnings.
HD Universe[]
Miguel Madrazo mentions that Juan Strickler is threatening to give the Madrazo Files to the DEA, rather than using the established HD Universe counterpart, the Drug Observation Agency (DOA). Rather than implying co-existence in each universe, this is presumably an oversight similar to the FIB/FBI textures in the 10th Anniversary edition of Grand Theft Auto III and in Grand Theft Auto: The Trilogy - The Definitive Edition
During the First Dose 6 - Off the Rails mission of the Los Santos Drug Wars update mentions the DEA. He states that precursor chemicals are on the government blacklist, also mentioning that you can't buy them without the DEA looking over your shoulder. Once again, this was likely an oversight on the part of Rockstar rather than them implying both the DEA and DOA co-exist.
