The Nintendo DS version of 101-in-1 Explosive Megamix garnered a mixed reception. Review aggregator Metacritic gave it a 46 out of 100, indicating "Generally mixed reviews."
Nintendo World Report gave it a 3 out of 10, stating "101-in-1 Explosive Megamix is the definitive bad budget title. Some of the mini-games are remotely fun, but others feel like a chore. With so many poor and forgettable games on the compilation, it's hard to recommend this one". Eurogamer rated it a 4 out of 10, stating "There is so little replay potential here, so little urge to top high scores or perfect shoddy make-do attempts, that completing each task feels more of a relief than an achievement". Nicholas Tan of Game Revolution gave the game a D- rating, saying "You're better off putting on a blindfold, walking into a rack of DS titles, and picking the game that your face lands on." Despite this, he praised the minigame variety.
The WiiWare version, by contrast, was met with a harsher critical reception. It received a 16 out of 100 on Metacritic, indicating "Overwhelming dislike."
IGN's Lucas M. Thomas gave the game a 2.5 out of 10, criticizing the controls. Additionally, Thomas also criticized the game for being a direct port of Nordcurrent's iPhone app 101-in-1 Megamix and its aforementioned Nintendo DS version, concluding with "[...] keep your cash held back from this 'value' option. It's a waste of money no matter how the math works out". Official Nintendo Magazine UK gave it 14 percent, noting "You'll find it tough to unlock all 101 games. Not that you'd want to bother because they're so abysmal, not even of the standard of your average Flash game".
The game, just like the Nintendo DS version, received a D- from Game Revolution. GamesRadar+ rated it a 4 out of 10, and Peter Willington of Nintendo Life gave it a 3 out of 10, stating that none of the minigames included were fun to play.
