• Mina Lynne Sparkles / Agent Xero (voiced by Mae Whitman) - a happy-go-lucky, naive and friendly teenage Modifyer agent who heads out for peril and adventure. It is known that her parents disappeared sometime before the series' start, although The Modifyers Movie reveals Xero's unnamed father was a Modifyer agent who was killed by Vain.
• Mole (voiced by Jeff Bennett) - Xero's shapeshifting robotic assistant who helps her with her missions by using his powers to aid her. He is usually the straight man to Xero's childish nature and crazy ideas.

Secondary

• Starlight (vocal effects also by Mae Whitman) - Xero's pet cat who occasionally follows her in her missions.
• Sparko (vocal effects by Dee Bradley Baker) - Xero's playful pet dog who occasionally follows her in her missions.
• Nigel Barbera / Agent Dos (voiced by James Arnold Taylor) - a funny Modifyer agent who is Xero's cousin and occasionally helps her with her missions as well as giving some advice to her.
