Giemsa stain (/ˈɡiːmsə/), named after German chemist and bacteriologist Gustav Giemsa, is used in cytogenetics and for the histopathological diagnosis of malaria and other parasites.
Uses
It is specific for the phosphate groups of DNA and attaches itself to regions of DNA where there are high amounts of adenine-thymine bonding. Giemsa stain is used in Giemsa banding, commonly called G-banding, to stain chromosomes and often used to create a karyogram (chromosome map). It can identify chromosomal aberrations such as translocations and rearrangements.
It stains the trophozoite Trichomonas vaginalis, which presents with greenish discharge and motile cells on wet prep.
Giemsa stain is also a differential stain, such as when it is combined with Wright stain to form Wright-Giemsa stain. It can be used to study the adherence of pathogenic bacteria to human cells. It differentially stains human and bacterial cells purple and pink respectively. It can be used for histopathological diagnosis of malaria and some other spirochete and protozoan blood parasites. It is also used in Wolbach's tissue stain.

How to do a hematology stain (Wright-Giemsa stain) Laboratory Tutorial Procedure
Wright-Giemsa Stains, also known as Romanowsky Stains, are stains used in hematology and cytological studies to differentiate cells in microscopic examinations of peripheral blood and bone marrow specimens.
These stains can also be applied to detect the presence of parasites in the blood when rapid results are needed, such as for the detection of the Plasmodium parasite that causes malaria.
There are a variety of Romanowsky stain types that apply the same principle. They include:
1. Giemsa stain
2. Wright and Wright-Giemsa stain
3. May-Grunwald stain
4. Leishman stain

Geimsa Stain -- for Plasmodium and Babesia Parasite Diagnostics
Alpha-Tec stains and staining reagents are specifically manufactured to minimize precipitate or sediment, making accurate identification easier and reducing the chance of false-positive diagnosis.
When producing thick smears, include Alpha-Tec’s Methylene Blue Phosphate Pre-Stain to preserve organisms, hemolyze red blood cells, and introduce color into parasite cytoplasm.

An easy mnemonic to remember Giemsa stain taking organisms is explained in this video. Trypanosomes, Plasmodium, Rickettsia, Borrelia, and Chlamydia are stained by Giemsa stain. These organisms are frequently tested in various competitive exams, like USMLE, MRCP, PLAB, AMC, DHA, NEET, and other examinations.
Giemsa stain is one of the most popular microscopic stains, commonly used in hematology, histology, cytology, and bacteriology for in vitro diagnostic use. Giemsa's stain is regarded as the standard diagnostic stain for malaria's plasmodium, and it is also the basic stain for classifying lymphomas in the Kiel classification.

Wright's stain is a hematologic stain that facilitates the differentiation of blood cell types. It is classically a mixture of eosin (red) and methylene blue dyes. It is used primarily to stain peripheral blood smears, urine samples, and bone marrow aspirates, which are examined under a light microscope.

Data: 2017
Resum: This project is for the students of the Biology Unit (Department of Pathology) from the degrees of Health Sciences. One of the objectives is to provide complementary multimedia material to prepare and better understand the laboratory practices and, on the other hand, to consolidate the knowledge that will be developed throughout it. To this end, this video describes in a virtual way the realization of a blood smear and the staining of its cellular components with Giemsa staining.

Giemsa stain is a mixture of Azure, Methylene blue, and Eosin dye. Giemsa stain is a differential stain that is used to variably stain the various components of the cells and it can be used to study the adherence of pathogenic bacteria to human cells. It differentially stains the human and bacterial cells and appeared as purple and pink colored bodies respectively.
The Giemsa stain is one of the best stains for malaria and other blood parasites and is also satisfactory as a routine bloodstain to stain the Peripheral blood smear for the examinations of blood film under the microscope.

Alpha-Tec stains and staining reagents are specifically manufactured to minimize precipitate or sediment, making accurate identification easier and reducing the chance of false-positive diagnosis.
When producing thick smears, include Alpha-Tec’s Methylene Blue Phosphate Pre-Stain to preserve organisms, hemolyze red blood cells, and introduce color into parasite cytoplasm.
For more information on these and other great products, visit http://www.alphatecsystems.com/parasitology/parasitology-stains-and-stain-sets/.
