In the season premiere ("The One After Vegas"), Ross and Rachel's marriage is established to be a drunken mistake, and although Ross lies and says he got the marriage annulled so that he won't have to be known as the "three divorces" guy, Rachel eventually finds out and they divorce several episodes later. Monica and Chandler decide to move into her apartment together, and Rachel moves in with Phoebe. Joey lands a role on a cable television series called Mac and C.H.E.E.S.E, where he stars alongside a robot. Ross gets a job lecturing at New York University, and starts dating one of his students, Elizabeth (Alexandra Holden). Phoebe and Rachel's apartment catches fire, and Rachel moves in with Joey while Phoebe moves in with Chandler and Monica. In the season finale, Chandler proposes to Monica.

The series finale aired on Sunday, May 23, 2010. The finale ran two-and-a-half hours starting at 9:00 pm; pushing the local news back a half-hour, followed by the previously announced post-finale special, Jimmy Kimmel Live: Aloha to Lost, at 12:05 am. The season continues the stories of the survivors of the fictional September 22, 2004 crash of Oceanic Airlines Flight 815 on a mysterious island in the South Pacific. The survivors must deal with two outcomes of the detonation of a nuclear bomb on the island in the 1970s. While the on-island story continues, "flash sideways" show a second timeline, in which Flight 815 never crashes. The season was released on DVD and Blu-ray on August 24, 2010 alongside a complete series boxset. Included in the special features was "The New Man in Charge," a previously unaired shortened episode that serves as an epilogue following the events of the finale.

In the Season 4 premiere, Ross decides to break up with Bonnie to get back together with Rachel, who writes him a letter. However, Ross is too tired to read the 18-page letter in the early hours of dawn, but his desire to get back with Rachel is such that he lied about reading it to her the next morning. Upon realizing that the letter had stated that Rachel is willing to reconcile with Ross only on condition that he accepts full responsibility for their previous break-up, he opposes. They break up again due to Rachel's continuous mention of the letter and Ross not being able to continue having Rachel think that he has taken responsibility for their first break up.
As Rachel journeys into motherhood, Monica and Chandler continue to try to have a child of their own but discover that they are unlikely to conceive when they go to a fertility specialist. Phoebe begins dating Mike Hannigan, who Joey set her up with at the last minute. Rachel moves out of Ross's apartment with Emma and moves back in with Joey when the two of them have a fight over them dating other people. Towards the end of the season Rachel finds herself developing romantic feelings for Joey after having a dream about the two of them together. While giving some paleontologists a tour of the city Ross finds himself attracted to one named Charlie who ends up going out with Joey. In the season finale, the group travels to Barbados to hear Ross give a keynote speech at a paleontology conference where Mike proposes to Phoebe and Charlie ends up kissing Ross; Joey seeing this decides to go ahead with a relationship with Rachel, after she had confessed her feelings for him.
