De adolescente, Marsalis inició su carrera marcada por Wayne Shorter y más tarde, pero de forma más destacada, por John Coltrane. En 1986 formó su propio grupo con Kenny Kirkland (piano), Bob Hurst (bajo) y Jeff Watts (batería). Su creciente interés por el jazz lo llevó a realizar sus primeros trabajos con grandes figuras como el trompetista Clark Terry y junto a su hermano Wynton con Art Blakey y The Jazz Messengers, tocando con Miles Davis en los últimos días de Davis.
