They were established in 1884 as Kempston Rovers F.C.. They joined the United Counties League in 1953. They won their first title in 1955–56; the following season Division Two and One were merged into one league and Kempston finished runners-up. In 1957–58, the club were crowned champions of the UCL. The league was split into two division again in 1961–62 and Kempston were placed in Division Two. Despite finishing tenth that season, they were promoted to Division One. In 1973, Division One was renamed the Premier Division and in 1974–75 the club were crowned champions again, and also reached the Fifth Round of the FA Vase. In 1978–79, the Kempston reached the Fourth Qualifying Round of the FA Cup, the furthest they have ever got in the competition. They repeated their record FA Vase run in 1980–81, reaching the Fifth Round. Rovers were relegated in 1983, but were promoted again in 1985 after winning Division One.
Until the 19th century Kempston was a mainly rural parish. It was one of the largest in Bedfordshire with an area of 5,025 acres (20km²) at the time of enclosure in 1804, and was in Redbournestoke Hundred. Historically there was no central village, but instead settlement was divided between a number of hamlets called "Ends", for example, Up End, Bell End, Wood End and Box End. Kempston's parish church, All Saints, was in Church End, which was not the largest end but is fairly central. In the 19th century East End, Bell End and Up End began to coalesce into a larger settlement. In 1870 developers began to attempt to develop land on the road from Kempston to Bedford under the name "Kempston New Town". Construction was slow at first, but the new district soon began to expand steadily and Kempston acquired a more urban feel. A permanent military presence was established in the town with the completion of Kempston Barracks in 1876.
