Autistic Spectrum Disorder (ASD) is a complex neurodevelopmental brain disorder characterized by two core behavioral symptoms, namely impairments in social communication and restricted/repetitive behavior. The molecular mechanisms underlying ASD are not well understood. Recent genetic as well as non-genetic animal models contributed significantly in understanding the pathophysiology of ASD, as they establish autism-like behavior in mice and rats. Among the genetic causes, several chromosomal mutations including duplications or deletions could be possible causative factors of ASD. In addition, the biochemical basis suggests that several brain neurotransmitters, e.g., dopamine (DA), serotonin (5-HT), gamma-amino butyric acid (GABA), acetylcholine (ACh), glutamate (Glu) and histamine (HA) participate in the onset and progression of ASD. Despite of convincible understanding, risperidone and aripiprazole are the only two drugs available clinically for improving behavioral symptoms of ASD following approval by Food and Drug Administration (FDA).
Till date, up to our knowledge there is no other drug approved for clinical usage specifically for ASD symptoms. However, many novel drug candidates and classes of compounds are underway for ASD at different phases of preclinical and clinical drug development. In this review, the diversity of numerous aetiological factors and the alterations in variety of neurotransmitter generation, release and function linked to ASD are discussed with focus on drugs currently used to manage neuropsychiatric symptoms related to ASD. The review also highlights the clinical development of drugs with emphasis on their pharmacological targets aiming at improving core symptoms in ASD. (See citation below)

Fetal Alcohol Syndrome (FAS) is one of five conditions within the fetal alcohol spectrum disorder (FASD), of which it is the most severe. All FASD conditions are due to prenatal exposure to alcohol (due to maternal consumption of ethanol or alcohol during pregnancy). In this lesson, we discuss the factor of alcohol in the pathophysiology of Fetal Alcohol Syndrome, including risk factors associated with alcohol consumption, the signs and symptoms of Fetal Alcohol Syndrome (facial morphology, brain abnormalities and skeletal abnormalities), how Fetal Alcohol Syndrome is diagnosis and how it is treated.

As a foster mother to children with FASD, Nora Boesem has seen the effects of alcohol first hand. However, FASD often goes undiagnosed and is creating a burden on us all.
Nora Boesem and her husband began fostering in 2011 and have fostered over 100 children for the state of SD and the Oglala Lakota Sioux Tribe. Nora realized there was no help for families and founded Roots to Wings in 2008, a non-profit to help advocate and support people living with FASD. In 2008 she also started a parenting group with the support of BMS in Rapid City, SD that still runs today and has now expanded into a beginners group and an advanced parenting group. In 2009 she was appointed to the Governor’s mental health board, joined the Chiesman Center for FASD and began speaking around the state of SD and around...

Natalie was born at 28 weeks as a result of fetal alcohol syndrome. Stephanie intended to find an adoptive family for her daughter upon birth, but after seeing Natalie for the first time instead found sobriety at rehab.

As a foster mother to children with FASD, Nora Boesem has seen the effects of alcohol first hand. However, FASD often goes undiagnosed and is creating a burden on us all.
Nora Boesem and her husband began fostering in 2011 and have fostered over 100 children for the state of SD and the Oglala Lakota Sioux Tribe. Nora realized there was no help for families and founded Roots to Wings in 2008, a non-profit to help advocate and support people living with FASD. In 2008 she also started a parenting group with the support of BMS in Rapid City, SD that still runs today and has now expanded into a beginners group and an advanced parenting group. In 2009 she was appointed to the Governor’s mental health board, joined the Chiesman Center for FASD and began speaking around the state of SD and around the US. Returning to school she earned her BS in psychology and is currently in her last year of her MSW at Arizona State University. In 2014 Nora joined BMS in a program she spearheads called Facing FASD.

It is vital for these families and the professionals who support them to understand the reasons behind any learning and behavioral challenges so they—together—can “try differently.” During this webinar, Barb Clark of the North American Council on Adoptable Children (NACAC), will share information about (1) fetal risk factors when illicit drugs, tobacco, and alcohol are used during pregnancy; (2) fetal alcohol spectrum disorders (FASDs) and the connection between brain impairment and behavior; (3) diagnoses under the FASD umbrella; (4) “red flags” for an FASD diagnosis; (5) comorbid conditions, including the impact of trauma; and (6) reframing and other strategies.

Rust trial: Expert witness ‘points gun at judge’
A witness in the trial of the weapons supervisor for Alec Baldwin's Rust film was reprimanded for pointing a gun at the judge.
Frank Louis Blair Koucky III, a firearms expert appearing for Hannah Gutierrez-Reed's defence, was given a stern warning by District Judge Mary Marlowe Sommer after brandishing an unloaded revolver for demonstration purposes.
"Everybody's nervous because you have not demonstrated to us they are unloaded," said the judge as Mr Koucky held the gun in the witness box. "So before you start showing us the weapons make sure they're unloaded, including that one you just touched."

When sweet Northern college kid Bill (Ralph Macchio) and his buddy Stan (Mitchell Whitfield) are picked up and thrown into the slammer in a hick Southern town, at first it looks like no big deal. Then they are informed that they are accused of murder. Penniless and without a single friend in the area, Bill decides to call his goofy cousin Vinny (Joe Pesci), who has somehow recently become a lawyer. Full of family feeling and bravado, Vinny, who has never tried a criminal case in his short life as a lawyer, rides south to defend his trusting relative. He's an expert motormouth and street-level logician from the wilder reaches of metropolitan New York, complete with a thick accent and the attitude to go with it. Otherwise, he's much less well qualified than your average public defender. When he arrives on the scene with his equally brassy girlfriend Lisa (Marisa Tomei), Bill is fairly sure he's going to be sentenced to death. His buddy Stan is even less confident of his legal representative, if that's possible, and the first thing Vinny has to do is to regain the consent of his clients to represent them. The local judge doesn't seem any too sympathetic to Vinny's verbal shenanigans either, and even the most optimistic supporter of the boys would begin to have doubts at this point -- and Vinny's no exception. With the insistent moral encouragement of his girlfriend, Vinny somehow accomplishes the impossible and wins grudging (if very irritated) respect from all concerned, for once studying as if his life depended on it.

FILM DESCRIPTION:
When sweet Northern college kid Bill (Ralph Macchio) and his buddy Stan (Mitchell Whitfield) are picked up and thrown into the slammer in a hick Southern town, at first it looks like no big deal. Then they are informed that they are accused of murder. Penniless and without a single friend in the area, Bill decides to call his goofy cousin Vinny (Joe Pesci), who has somehow recently become a lawyer. Full of family feeling and bravado, Vinny, who has never tried a criminal case in his short life as a lawyer, rides south to defend his trusting relative. He's an expert motormouth and street-level logician from the wilder reaches of metropolitan New York, complete with a thick accent and the attitude to go with it. Otherwise, he's much less well qualified than your average public defender. When he arrives on the scene with his equally brassy girlfriend Lisa (Marisa Tomei), Bill is fairly sure he's going to be sentenced to death. His buddy Stan is even less confident of his legal representative, if that's possible, and the first thing Vinny has to do is to regain the consent of his clients to represent them. The local judge doesn't seem any too sympathetic to Vinny's verbal shenanigans either, and even the most optimistic supporter of the boys would begin to have doubts at this point -- and Vinny's no exception. With the insistent moral encouragement of his girlfriend, Vinny somehow accomplishes the impossible and wins grudging (if very irritated) respect from all concerned, for once studying as if his life depended on it.

A witness in the trial of the weapons supervisor for Alec Baldwin's Rust film was reprimanded for pointing a gun at the judge.
Frank Louis Blair Koucky III, a firearms expert appearing for Hannah Gutierrez-Reed's defence, was given a stern warning by District Judge Mary Marlowe Sommer after brandishing an unloaded revolver for demonstration purposes.
"Everybody's nervous because you have not demonstrated to us they are unloaded," said the judge as Mr Koucky held the gun in the witness box. "So before you start showing us the weapons make sure they're unloaded, including that one you just touched."
Mr Koucky then drew a gasp from the public gallery at the courthouse in Santa Fe, New Mexico on Monday when he appeared to point the muzzle of the revolver in the judge's direction in a bid to prove it was unloaded.
