Amphetamine is a stimulant drug used mainly in the treatment of ADHD. Several popular drugs, like Adderall, primarily contain amphetamine.
Although we don’t have a full understanding of the effects of amphetamine on the brain, there are some aspects of its pharmacology that are well established. Amphetamine binds to the transporter proteins for monoamines like dopamine, norepinephrine, and serotonin and then can be taken up into neurons via these transporter proteins. Once inside the neuron, amphetamine disrupts the storage of monoamines in synaptic vesicles. One way it does this is by inhibiting
a protein called vesicular monoamine transporter 2, or VMAT2, whose normal role is to transport monoamines into vesicles. Inhibition of VMAT2 leads to higher levels of these neurotransmitters in the neuron.
Through a mechanism or mechanisms that are still not fully understood, amphetamine is then able to cause the monoamine transporter proteins to run in reverse, leading to the increased release of monoamines---especially dopamine and norepinephrine---and increased levels of these neurotransmitters in the synaptic cleft. These elevated monoamine levels can have various effects on different parts of the central nervous system. Increased dopamine levels in the reward system, for example, may contribute to the reinforcing effects of amphetamine use.
Although the primary action of amphetamine is to cause the increased release of monoamines, there are various other mechanisms that may contribute to the effects of the drug. For example, amphetamine also competes with monoamines for reuptake into the neuron, in effect inhibiting reuptake. And it may inhibit the activity of monoamine oxidase, an enzyme that metabolizes monoamines. The effect of both of these actions is to additionally increase levels of monoamines in the neuron and the synaptic cleft.

The origin of Amphetamines:
It all starts about 5000 years ago in China. There grew a plant called Ephedra, which contained the amphetamine-like stimulant ephedrine. Which was at that time already used by the local population for its effects.
If we fast forward to about 1885, ephedrine was for the first time isolated form the plant. This was done in Japan, after it was studied for its medicinal properties. In the years to come, scientist were also able to synthesize Amphetamine, and from 1933 onwards it was sold as a drug to potentially treat a wide variety of medical conditions: narcolepsy, obesity, low blood pressure, chronic pain and ADHD in children.
Interestingly, up until, the 1960-70 it’s addictive properties were mostly unknown. But, from that point onwards, governments started to place strict controls on the sale of amphetamines.
What actually are Amphetamines?
There are 4 common types of amphetamines;
- Amphetamine: called speed, uppers, goey, louee or whiz.
- Dextroamphetamine (ADHD medicine used illegally): dexies, kiddie-speed, pep pills, uppers; black beauty (when combined with amphetamine).
- Methamphetamine (crystal solid form): crystal, meth, base, d-meth, fast, glass, ice, speed, whiz, pure, wax.
- Methamphetamine (liquid form): leopard's blood, liquid red, ox blood, red speed.
These types of amphetamines, come as pills, capsules, powder, paste, crystal and liquid, and therefore can be used by swallowing, dabbing onto the gums, smoking, snorting and injecting.
How does it work?
They work by increasing the dopamine and noradrenaline levels in multiple pathways of your central nervous system. Both are neuromodulatory molecule, which effect the reward centrum in your brain, your thinking, mood and movement.
The short term effects:
- Burst of energy (making you talkative, restless and excited), feelings of euphoria, feelings as if your thinking is extremely clear, accelerated heart rate and breathing rate, high blood pressure, dry mouth, jaw clenching, sweating, dilated (lage) pupils), loss of appetite, increased libido and nose bleeds (if you snort it).
Risks of using Amphetamines:
- A binge crash might occur. This can take a few days. You might experience; extreme exhaustion, requiring a lot of sleep, headaches, muscles twitches, irritability, mood swings, depression, paranoia, hallucinations, confusion and there is a risk of using other drugs to cope with the “crash”
- Furthermore, there is always the risk of an overdose. It might occur when taking a too strong batch, using a large amount or mixing it with other drugs or alcohol.
Long term risks:
Psychosis, extreme weight loss, malnutrition, dental problems, regular infections, mood swings, panic attacks, anxiety, depression, drug dependence, aggression, heart and kidney problems and financial, work, study, legal and relationship problems.
In a nutshell, as with any serious addiction, it could impact and destroy someone's whole life.

- Depending on your experience, the way of using it and the strength of the speed, 5 to 40 milligrams is a usual dose.
RISKS:
- Overheating (hypothermia). Make sure to rest enough in a cooler area and drink up to 2 glasses of water in one hour;
- Increased risk of heart problems;
- Brain damage;
- Addiction;
- Physical problems;
- Hangover.

AMPHETAMINE
Kafka, a straight fitness trainer meets Daniel, a passionate executive who happens to be gay. The young men fatefully fall in love and believe that their love can bridge anything, even their difference in sexuality and Kafka’s drug habits. Daniel does not regret his love for Kafka, who tries to love him back against his nature. But a dreadful memory from Kafka’s past render it most difficult. Im Chinesischen besitzt das Wort "Amphetamin" noch eine weitere Bedeutung, nämlich: "Ist dies nicht sein Schicksal?". Von ihm wird in diesem leidenschaftlichen Drama ein junger Fitnesstrainer namens Kafka eingeholt. Kafka -- so schön wie Michelangelos David -- arbeitet nebenher als Kampfsportkünstler und Botenjunge. Nur mithilfe mehrerer Jobs kann er seine kranke Mutter finanziell unterstützen.
Eines Nachts -- es ist dieselbe Nacht, in der er sich von seiner Freundin May getrennt hat -- lernt Kafka den hübschen jungen Finanzmanager Daniel (THOMAS PRICE) kennen. Daniel ist aus Australien nach Hongkong zurückgekehrt. Daniel umwirbt Kafka sehr heftig, und Kafka gibt diesen Avancen nach -- weil er einsam ist und sich nach Liebe sehnt. Die beiden Männer sind davon überzeugt, dass ihre Liebe alles überbrücken kann, was sie voneinander unterscheidet, ihre gesellschaftliche Stellung ebenso wie die Drogensucht.
Daniel hat keinen Grund, seine bedingungslose Liebe zu Kafka zu bedauern. Kafka versucht sie zu erwidern, indem er diese Art der Liebe, die ihm nicht in die Wiege gelegt wurde, akzeptiert. Doch dann kompliziert eine furchtbare Erinnerung an die Vergangenheit die Situation. Wenn schon die Sucht nach Amphetaminen tödlich ist, dann ist die Sucht nach Liebe noch viel schicksalhafter. Der lange, schwierige Weg führt Kafka schließlich zu sich selbst.
Seinen Film über Sex, Drogen und die wahre Liebe möchte Scud jenen Menschen gewidmet wissen, die in der Liebe den einzigen Zweck ihres Daseins finden und gleichwohl die Liebe ihres Lebens verloren haben.
AMPHETAMIN ist nach PERMANENT RESIDENCE der zweite Teil einer autobiografischen Trilogie von Scud.
