Hello everyone,

I was wondering if anyone experienced difficulties with crossing different reporter strains into the strain MQ989 (isp-1(qm150) IV; ctb-1(qm189))? I have been trying to cross different transcriptional reporters and mutants into this genetic background, but for a large number of crosses I attempted, I keep getting the progeny that is heterozygous for these mutations or to a low percentage wild-type, but I fail to get homozygous worms. I do not expect that any of these transgenes would be lethal in this genetic background, so I am not sure what could be the reason for this. I singled hundreds of worms already.

Has anyone experienced this before?
If yes, do you have any suggestions?
