Kloniranje i eksresija
[uredi | uredi izvor]Ovaj gen kodira transmembranski i izlučeni protein sa karakteristikamatransmembranskih proreina tipa 1a. Nalazi se u perijedarnom Golgijevom obrascu i smatra se da je uključen u procese hematopoeze i/ili imunskog sistema.[6]
Dužina polipeptidnog lanca je 248 aminokiselina, a molekulska težina 27.039[7].
Analizom DNK-hipersenzibilne regije, otprilike 5 kb uzvodno od gena CD7 na 17q25.2-q25.3, Slentz-Kesler et ak. (1998) klonirali su SECTM1 cDNK, koju su nazvali K12. Northern blot analiza otkrila je da se K12 eksprimira kao iRNK od 1,8 kb u mnogim testiranim tkivima, sa najvišim nivoom ekspresije u leukocitima periferne krvi. Predviđeni protein K12 od 248 aminokiselina ima hidropatske karakteristike transmembranskog proteina tipa 1a, koji je definiran vanćelijskim N-krajem i odcijepljenim signalnim peptidom. Autori su pokazali da se protein K12 biohemijski ponaša kao integralni membranski protein. Koristeći imunofluorescenciju, lokalizirali su K12 na Golgijevom aparatu. Western blotiranje pokazalo je da protein K12 migrira kao klaster traka na 27 kD. Pored toga, ćelije luče približno 20 kD rastvorljivog oblika K12. Utvrdili su da gen K12 sadrži četiri egzona i da se proteže približno na 14 kb.[8]
