Maguire was born in Newport News, Virginia, and, as a teenager, worked as a strolling troubadour in Williamsburg, Virginia, before going on to study opera at the Oberlin Conservatory and the University of Michigan. One of his sisters is Linda Maguire, a renowned classical and operatic vocalist. He worked as a stockbroker for several years before making his Broadway debut in 1987 with Les Misérables. He appeared in the films L.A. Pictures, Cadillac, Go Fish, The Deep End Of The Ocean, Busted, and Where The Day Takes You, as well as in a variety of television shows.
In his first year as Wigan's coach Maguire won the 2010 Super League Grand Final. In his second year he won the 2011 Challenge Cup and was signed by the Rabbitohs. In his first year as an NRL head coach he took the Rabbitohs deep into the 2012 finals series. In 2013 the Rabbitohs finished the regular season in second place and in 2014 they won their first premiership in 43 years.
He holds the record as the youngest player to have played senior football at Richmond, being only 15 years and 328 days old upon debut in Round 1 of the 1910 season. Despite this, he was the club's leading goalkicker in his debut year. He also gained some notoriety as a welterweight boxer between 1912 and 1915.
Maguire was a well-known publican in Melbourne, operating the Bull and Mouth Hotel in Melbourne, and after 1932, Bellevue Hotel in Brisbane. He was father to five daughters; sometimes known as "the fabulous Maguires." Two daughters married members of the English nobility and Mary Maguire was briefly a Hollywood and British film actress in the late 1930s. Maguire served in the British Army in World War Two and died in England in 1950.
