Pérez is one of 11 siblings. By the time Pérez finished high school, Manny knew he wanted to be an actor, he moved to New York where he majored in drama at Marymount Manhattan College, graduating in 1992.
He has also studied at Ensemble Studio Theatre and is a member of the LAByrinth Theatre Company, in New York City.
After his performance on the film Washington Heights which he also co-wrote and produced, Manny garnered praise from many critics.
The New York Times stated "Mr. Perez has charisma to burn." He won the 2002 Best Actor Award at The Milan International Film Festival in Italy for his performance in Washington Heights.
Pérez was the first ever member of his family to get a university degree.
Born in 1914 in Morenci in Arizona, in 1917 his family moved to Los Angeles where Perez later attended high school. An athlete, he took part in football, baseball and track. Two years after graduating he was hired by Leon Schlesinger as a trainee animator. On 17 April 1938 he married Connie (née Perez).

Manuel 'Manny' Perez (1914 - 1981) was a Mexican American animator whose career started at Warner Bros. under the supervision of Friz Freleng. Later on, he would work for Bill Melendez Productions, DePatie-Freleng Enterprises, Hanna-Barbera, and Ralph Bakshi with "Fritz the Cat", "Heavy Traffic" and "The Lord of the Rings" (1978).
Perez's animation is somewhat jerky (like Gerry Chiniquy's), but also solid at the same time.
