The optical microscope, often referred to as light microscope, is a type of microscope which uses visible light and a system of lenses to magnify images of small samples. Optical microscopes are the oldest design of microscope and were possibly invented in their present compound form in the 17th century. Basic optical microscopes can be very simple, although there are many complex designs which aim to improve resolution and sample contrast.
The image from an optical microscope can be captured by normal light-sensitive cameras to generate a micrograph. Originally images were captured by photographic film but modern developments in CMOS and charge-coupled device (CCD) cameras allow the capture of digital images. Purely digital microscopes are now available which use a CCD camera to examine a sample, showing the resulting image directly on a computer screen without the need for eyepieces.

This arrangement in the microscope causes differences in light intensity while passing through the specimen so that denser parts alter the path of light more than the less dense cytoplasmic region. The image formed, therefore, has a varying contrast for different regions.

Ultraviolet Microscope:
Casperson invented the ultraviolet microscope where the illuminating source is ultraviolet rays from mercury or iodine-Quartz lamp. Image is captured on photographic film.

Fluorescence microscope:
It is a modification of an ultraviolet microscope. Fluorescent dyes absorb UV light and emit longer-wavelength light, which is used to stain specific macromolecules of cells. The fluorescence microscope makes the use of two filters, one filter permits only short-wavelength light to pass through the specimen for observation. Another one is used to block light emitted by fluorescent dye. It is used to study photochemical reactions within living cells.

Confocal microscope:
In this case, a laser beam is focused to scan the: specimen, which produces a clear image of one plane of the specimen, while other planes of the specimen are excluded and don’t blur the image.

NUMBER 1.
Dark-field Microscope
It was invented by Zsigmondy. In a darkfield microscope, a special condenser lens is used with an opaque disc at the center, so that direct rays don’t enter the objective lens. Only light scattered by the specimen enters the objective lens to form a bright image against a dark background. The Darkfield microscope does not have a good resolution. It is used in microbiology and in autoradiography.
NUMBER 2.
Phase-Contrast microscope:
It is a modified light microscope developed by Frederick Zernicke. In this microscope, an annular diaphragm is issued below the condenser lens and a transparency phase plate above the objective. This arrangement in the microscope causes differences in light intensity while passing through the specimen so that denser parts alter the path of light more than the less dense cytoplasmic region. The image formed, therefore, has a varying contrast for different regions.
NUMBER 3.
Ultraviolet Microscope:
Casperson invented the ultraviolet microscope where the illuminating source is ultraviolet rays from mercury or iodine-Quartz lamp. Image is captured on photographic film.
NUMBER 4.
Fluorescence microscope:
It is a modification of an ultraviolet microscope. Fluorescent dyes absorb UV light and emit longer-wavelength light, which is used to stain specific macromolecules of cells. The fluorescence microscope makes the use of two filters, one filter permits only short-wavelength light to pass through the specimen for observation. Another one is used to block light emitted by fluorescent dye. It is used to study photochemical reactions within living cells.
NUMBER 5.
Confocal microscope:
In this case, a laser beam is focused to scan the: specimen, which produces a clear image of one plane of the specimen, while other planes of the specimen are excluded and don’t blur the image.

Do digital microscope specs match reality?
This video has been a year in the making. Last June, 2022, I purchased a variety of six different digital microscopes that I saw on the Amazon website. All of them boasted extremely high magnification numbers, so I was immediately skeptical of their capabilities. A microscope cable of 1000x magnification for only $10? That sounds way too good to be true…
For each of the six microscopes, I will take photographs at their highest magnification setting and compare them against my metallurgical microscope’s photos. In its current configuration for this video, my ME580 metallurgical microscope is capable of a maximum of 500x magnification. I’ll use two objects for these photos; a microscope calibration slide, and a silicon chip that happens to have a doodle of Sonic on it.

I would highly recommend against purchasing these cheaper digital microscopes if you were hoping to look at silicon chips up close. Even though they may be more expensive, there are many better options available.

In case you are wondering, this video took me 110 hours to make between testing microscopes, recording footage from all of them, and editing. These videos take a lot longer to make than people realize.
The MS-040, 40-400X Microscope is a entry level, Biological Microscope that is ideal for a student or family. The Microscope has a monocular head, at 45 degree angle for comfortable viewing, and is rotatable 360 degrees for easy sharing of the specimen view. Illumination is provided by two bright white LED sources (transmission and reflection) which allow viewing of wide range of transparent to opaque specimens. It features a filter wheel with 5 color options. The included AC power adaptor allows for use internationally without the need for a voltage transformer.
